Ⅰ. Overview

YPD Agar (YPD or YEPD) and YPD Broth are nutrient rich media, non-selective media used to maintain and propagate saccharomyces cerevisiae and other yeasts in various molecular microbiology procedures.

This medium promotes the vigorous growth of wild strains as well as mutant strains of all kinds of yeasts.

YEPD === Yeast extract peptone dextrose

Ⅱ. Preparation / composition

Suspend the components, dehydrated powder, in water (65 grams in 1000 ml of purified/distilled water). The medium is boiled for a few seconds until the ingredients are completely dissolved.

Cool to 47°C and pour into Petri-dishes. The final pH should be 7.5 + 0.2.


Ⅲ. Principle

Yeast can grow in media containing salts and glucose. However, the presence of peptone and yeast extract promotes faster growth.

  • Yeast extract provides B-complex vitamins and contains all the amino acids necessary for growth.
  • Peptone acts as a source of nitrogen, vitamins and minerals.
  • Dextrose serves as a carbon source.
